Whole Transcriptome Sequencing (WTS).
The analysis methodology is based on the creation of complementary DNA (cDNA) in order to generate a transcriptome library for research purposes. To apply NGS technologies to transcriptome analysis it has been established that this is more complex and dynamic of what initially had been described, including a wide variety of RNA molecules (short RNA, RNA …).
All the transcriptome sequencing is the more complete, cost-effective method for identification and quantification of RNA from different types of source material. Currently the Ion PI ™ chip – depending on desired sequencing coverage – allows analysis of one to four full transcriptomas chip (assuming more than 10 million sequences mapped sample), thanks to the obtaining of between 60-80 million sequences that allow the detection of low abundance transcripts and rare types of RNA, among other applications.
Another approach is the use of targets in the study of RNA, enabling the analysis of hundreds of samples (up to 384) in a single process of sequencing. Genetracer Biotech has taken advantage of the workflow of the AmpliseqTM Ion to be applied in the study of targets on RNA. This technology allows to choose among more than 20,000 specific genes using a tool online. There are currently also available specific panels for cancer and the apoptotic pathways. The technology of the AmpliseqTM together with the design of primers allows to analyze up to 300 genes selected by the researcher in a single reaction based on minimum quantities of RNA, from 500 pg.
Altogether, the analysis of the differential expression of RNA genome-wide or specific targets for specific objectives offers researchers a better understanding of biological pathways and molecular mechanisms that regulate cell fate, the development and progression of the disease.
